Super Broth: 3.2% peptone, 2% yeast extract, and 0.5% NaCl
Superbroth
deionized H2O, 950 ml tryptone, 32g yeast extract, 20g NaCl, 5g NaOH (1N), 5 ml
To prepare 1 liter of superbroth, mix the reagents in 950 ml H2O and shake until the solutes have dissolved. Adjust the volume of the solution to 1 liter with deionized H2O. Sterilize by autoclaving for 20 minutes at 15 psi (1.05 kg/cm 2) on liquid cycle.
