M9

M9 salts x 10 500 mL

• 35 g Na2HPO4•7H2O
• 15 g       KH2PO4
• 5 g        NH4Cl
• 2.5 g      NaCl

Use tap water, which includes many trace metal ions, to bring to 500 mL. Auoclave.

Before use add the following to 1/2 liter of 1x M9.

• 50 mL M9 salts
• 1 ml      1M MgSO4 (Filter)
• 2.5 ml     50% Carbon source (glucose or glycerol)
• 0.1 ml     0.5% Thiamine, Vitamin B1 (Filtered)

Add 100 uL of 1M CaCl2 (sterile, 7.35 g in 50 mL)

Adding sugar prior to autoclaving will cause sugar to turn brown or black.
